Just found a solution.  The trick is to use a reg exp that excludes a 
plain 'dir'

Here is the config part

<Directory /usr/www/dir>
  DirectoryIndex home
</Directory>

<Location ~ ^/dir/.+$>
  SetHandler mod_python
  ...
</Location>
